En utilisant plus de 1,4 pe\u0301taoctet de donne\u0301es provenant de l\u2019imagerie par microscopie e\u0301lectronique, l\u2019e\u0301quipe du professeur Jeff Lichtman est parvenue a\u0300 reconstruire nume\u0301riquement un minuscule segment cubique du cerveau humain.<\/p>\n<p>Bien que mesurant un millime\u0300tre de co\u0302te\u0301, ce fragment microscopique de cortex humain abrite pas moins de 57 000 cellules, 150 millions de synapses et 230 millime\u0300tres de veines ultrafines. Fruit de pre\u0300s de dix ans de travaux, il s\u2019agit de la reproduction la plus e\u0301tendue et la plus de\u0301taille\u0301e du cerveau humain a\u0300 ce jour, avec une re\u0301solution allant jusqu\u2019aux synapses, les e\u0301le\u0301ments cle\u0301s permettant la transmission des signaux entre les neurones.<\/p>\n<p><b>La cartographie d\u2019un connectome<\/b><\/p>\n<p>\u00ab Le mot \u2018fragment\u2019 est ironique \u00bb, pre\u0301cise Jeff Lichtman, dans une e\u0301tude parue dans Science. \u00ab Un te\u0301raoctet est, pour la plupart des gens, gigantesque, mais un fragment de cerveau humain \u2013 juste un tout petit morceau de cerveau humain \u2013 repre\u0301sente quand me\u0302me des milliers de te\u0301raoctets. \u00bb<\/p>\n<p>Puisque le cerveau humain et les neurones restent un domaine quasi inexplore\u0301, Lichtman et ses colle\u0300gues se sont attele\u0301s a\u0300 cartographier ce qu\u2019ils appellent un \u00ab connectome \u00bb, soit une repre\u0301sentation de\u0301taille\u0301e du ca\u0302blage ce\u0301re\u0301bral qui pourrait e\u0301clairer les dysfonctionnements e\u0301ventuels dans les maladies neurologiques.<\/p>\n<p>Alors que leur objectif principal est la reproduction comple\u0300te du cerveau de la souris, l\u2019application de techniques similaires a\u0300 des segments du cerveau humain pourrait conside\u0301rablement acce\u0301le\u0301rer la recherche me\u0301dicale.<\/p>\n<p><b>Google implique\u0301 dans cette de\u0301couverte<\/b><\/p>\n<p>Derrie\u0300re cette prouesse, il y a aussi l\u2019apport de l\u2019e\u0301quipe de recherche Connectomics de Google. La reconstruction, base\u0301e sur un e\u0301chantillon de cerveau humain pre\u0301leve\u0301 lors d\u2019une intervention chirurgicale, a ne\u0301cessite\u0301 des techniques avance\u0301es telles que la microscopie e\u0301lectronique a\u0300 coupe en se\u0301rie a\u0300 haut de\u0301bit.<\/p>\n<p>Cette me\u0301thode a permis de ge\u0301ne\u0301rer des donne\u0301es massives, qui ont ensuite e\u0301te\u0301 analyse\u0301es a\u0300 l\u2019aide d\u2019algorithmes spe\u0301cialise\u0301s, aboutissant a\u0300 une reconstruction 3D de\u0301taille\u0301e de chaque cellule et processus dans l\u2019e\u0301chantillon aligne\u0301.<\/p>\n<p>\u00ab Nos collaborateurs ont ge\u0301ne\u0301re\u0301 l\u2019ensemble de donne\u0301es a\u0300 l\u2019aide d\u2019un e\u0301chantillon de tissu ce\u0301re\u0301bral du lobe temporal ante\u0301rieur gauche qui a e\u0301te\u0301 retire\u0301 lors d\u2019une intervention chirurgicale au cerveau sur une personne e\u0301pileptique \u00bb explique Viren Jain, chercheur scientifique et responsable technique chez Google. \u00ab L\u2019e\u0301quipe de Lichtman a utilise\u0301 un microscope e\u0301lectronique a\u0300 balayage multifaisceau pour recueillir des images haute re\u0301solution de plus de 5 000 tranches de tissu, chacune d\u2019une e\u0301paisseur d\u2019environ 30 nanome\u0300tres.
